**Tusamitamab ravtansine + pembrolizumab + cisplatin** is a combination regimen composed of three agents: - *Tusamitamab ravtansine* is an antibody-drug conjugate targeting carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5), delivering DM4 (ravtansine) cytotoxic payload to CEACAM5-expressing tumor cells, resulting in microtubule inhibition and cell death. - *Pembrolizumab* is a humanized monoclonal antibody targeting PD-1 (programmed death-1), blocking the immune checkpoint pathway to activate T-cell mediated anti-tumor immunity. - *Cisplatin* is a platinum-based chemotherapy that induces DNA crosslinking and damage, resulting in apoptosis of rapidly dividing cells. This combination is investigational in oncology settings where synergistic immunogenic and cytotoxic effects are pursued, notably for solid tumors with high CEACAM5 expression and immunogenic profiles. Concurrent use aims to capitalize on targeted cytotoxicity (ADC), immune checkpoint blockade, and chemotherapy-sensitization for enhanced anti-tumor efficacy.
