Drug intelligence / Profile preview
This is a combination regimen consisting of the following: - **Tusamitamab ravtansine**: an antibody-drug conjugate (ADC) targeting CEACAM5 (carcinoembryonic antigen-related cell adhesion molecule 5). It comprises a monoclonal antibody linked to the cytotoxic agent DM4 (ravtansine, a maytansinoid), enabling selective delivery to CEACAM5-positive cancer cells, particularly in non-small cell lung cancer (NSCLC) and other solid tumors. - **Sintilimab**: a PD-1 (programmed death-1) immune checkpoint inhibitor, which is a fully human IgG4 monoclonal antibody designed to enhance antitumor immune response by blocking the interaction between PD-1 and its ligands. - **Carboplatin**: a platinum-based chemotherapy agent that induces DNA crosslinking, leading to apoptosis in rapidly dividing cancer cells. - **Pemetrexed**: a folate antimetabolite chemotherapy that inhibits multiple folate-dependent enzymes essential for DNA synthesis. The regimen is under investigation in Phase 2 clinical trials for the treatment of advanced or metastatic non-squamous non-small cell lung cancer (NSQ NSCLC) with CEACAM5-positive expression[1][3][4][5][6].
