Tusamitamab ravtansine is an investigational antibody-drug conjugate (ADC) that targets carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5), a glycoprotein highly expressed in non-small cell lung cancer (NSCLC), gastric cancer, and other solid tumors. It consists of a humanized monoclonal antibody linked to the cytotoxic agent DM4, designed to deliver targeted chemotherapy directly to CEACAM5-expressing tumor cells[1][3][4]. Sintilimab is a fully human IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), acting as an immune checkpoint inhibitor to enhance antitumor immune responses; it is approved in China for Hodgkin's lymphoma and under investigation for various solid tumors[6]. Carboplatin or cisplatin are platinum-based chemotherapeutic agents that induce DNA crosslinking and apoptosis, while pemetrexed is an antifolate chemotherapy inhibiting folate-dependent enzymes essential for DNA synthesis.
