TUTI-16 is a fully synthetic, self-adjuvanting lipopeptide vaccine designed as a universal HIV-1 Tat epitope vaccine. Its primary mechanism is to induce anti-Tat antibodies that block the function of circulating Tat, an HIV-1 encoded protein secreted by infected cells. The Tat protein activates CD4+ T cells and promotes HIV replication and sustained viremia. By generating antibodies against Tat, TUTI-16 aims to inhibit this process, potentially reducing viral load and immune suppression associated with chronic HIV infection. Clinical studies have shown that TUTI-16 is safe and can elicit robust antibody responses in healthy individuals and ART-controlled patients; however, immunogenicity was limited in treatment-naïve subjects at higher doses. The drug has been investigated as a therapeutic vaccine for controlling HIV viremia without continuous antiretroviral therapy[1][2][4][5].
