Overview

Tuvatexib (formerly known as VDA-1102) is a first-in-class, small molecule anti-neoplastic agent developed for topical treatment of actinic keratosis (AK) and cutaneous T-cell lymphoma (CTCL). It acts as a selective modulator of the voltage-dependent anion channel/hexokinase 2 (VDAC/HK2) complex in cancer cells. By allosterically binding to hexokinase 2, tuvatexib disrupts its interaction with VDAC on the mitochondrial membrane, leading to inhibition of glycolysis and induction of apoptosis specifically in malignant cells while sparing healthy tissue. This mechanism reverses the Warburg effect seen in cancer metabolism and reactivates immune responses against tumor cells. Tuvatexib is being developed by Vidac Pharma as both a topical ointment for skin cancers and an injectable formulation for solid tumors[1][4][5][6][8].
