TVB-2023 is a small molecule inhibitor of Fatty Acid Synthase (FASN), an enzyme that catalyzes the de novo synthesis of long-chain fatty acids. Developed by Sagimet Biosciences (formerly 3-V Biosciences), TVB-2023 is designed to disrupt the metabolic adaptations of cancer cells, particularly those that rely on lipid synthesis for survival and proliferation. In preclinical models of pancreatic ductal adenocarcinoma (PDAC), TVB-2023 has shown enhanced efficacy under high glucose conditions, which trigger the upregulation of lipid-synthesizing enzymes and increase cellular dependency on the FASN pathway. This metabolic rewiring sensitizes cancer cells to FASN inhibition, making TVB-2023 a potential therapeutic candidate for hyperglycemic or metabolically active tumors.
