TVD3-003 is a proprietary transdermal cholecalciferol (vitamin D3) patch formulation being developed by eptivA Therapeutics (under license from To Better Days) for the treatment of rare chronic pain conditions. Cholecalciferol acts as a prohormone that is metabolized in vivo to its active form, calcitriol, which subsequently binds to and activates the Vitamin D receptor (VDR). Activation of VDR is thought to suppress hyperactivity in nociceptor pain receptors and reduce neurogenic inflammation, thereby alleviating neuropathic and chronic pain. TVD3-003 is designed to provide continuous, sustained release of cholecalciferol with an improved half-life profile compared to standard oral administration, minimizing systemic side effects and avoiding gastrointestinal first-pass metabolism.
