twist1-sirna is a small interfering RNA (siRNA) designed to specifically silence the expression of the Twist-related protein 1 (TWIST1) gene, a basic helix-loop-helix transcription factor implicated in epithelial-mesenchymal transition (EMT), metastasis, and chemoresistance in various cancers. By targeting and degrading TWIST1 mRNA, twist1-sirna reduces TWIST1 protein levels, thereby reversing drug resistance phenotypes and increasing chemosensitivity to agents such as cisplatin and taxol. Preclinical studies have demonstrated that delivery of twist1-sirna via nanoparticles or dendrimers leads to significant knockdown of TWIST1 expression in cancer cells both in vitro and in vivo, resulting in reduced tumor burden and enhanced response to chemotherapy[3][4][5]. The approach is under investigation primarily for ovarian cancer but has potential applications across multiple solid tumors where TWIST1 drives malignancy.
