Overview

TWS119 is a potent, selective, synthetic, small-molecule inhibitor of glycogen synthase kinase-3 beta (GSK-3β), classified as a 4,6-disubstituted pyrrolopyrimidine. It has been widely used in biomedical research as an activator of the canonical WNT/β-catenin signaling pathway by inhibiting GSK-3β. TWS119 has shown activity in promoting neurogenesis, enhancing neuronal differentiation following hypoxic-ischemic brain injury, protecting neurons against oxygen-glucose deprivation-induced injury, modulating immune cell function, and exerting anti-inflammatory effects via the inhibition of GSK-3β. It has also been investigated in cancer research for effects on immune cell proliferation and function, and proposed as a candidate for neurodegenerative disorders and mild cognitive impairment, but is not approved for clinical use[1][2][3][4][5][6][7][8][9].
