TX000045 is a novel Fc-relaxin fusion protein developed by Tectonic Therapeutic for the treatment of pulmonary hypertension secondary to heart failure with preserved ejection fraction (PH-HFpEF) and congestive heart failure. It acts as a selective agonist of relaxin receptor 1 (RXFP1), a G protein-coupled receptor. By activating RXFP1, TX000045 mimics the effects of endogenous relaxin hormone, leading to vasodilatory, anti-fibrotic, and anti-inflammatory activities. Relaxin is naturally upregulated during pregnancy to reduce vascular resistance and increase cardiac output; similarly, TX000045 aims to improve hemodynamics in patients with PH-HFpEF by reducing systemic and pulmonary vascular resistance and increasing cardiac output. The drug has optimized pharmacokinetics and biophysical properties for clinical use[1][3][5][6].
