TX103 is an autologous chimeric antigen receptor T-cell (CAR-T) therapy developed by Beijing Tiantan Hospital for the treatment of recurrent glioblastoma. This cell-based biologic therapy involves genetically modifying a patient's own T cells to express a chimeric antigen receptor that specifically targets B7-H3 (also known as CD276), a protein highly expressed on glioblastoma cells but with limited expression in normal tissues. The therapy is administered intracranially through an Ommaya device placed into the tumor resection cavity or ventricular system, allowing direct delivery to the brain tumor site and bypassing the blood-brain barrier. TX103 is currently being evaluated in a Phase 1 dose-escalation clinical trial (NCT05241392) to assess its safety, tolerability, and preliminary effectiveness, with early results indicating a manageable safety profile and signs of clinical response.
