TX803 is an investigational small-molecule anticancer agent that selectively targets a truncated form of retinoid X receptor alpha (tRXRα), a mutant nuclear receptor implicated in oncogenic signaling. By binding tRXRα and inducing its tetramerization, TX803 disrupts tRXRα-dependent activation of the PI3K/AKT survival pathway, thereby inhibiting cancer cell growth while sparing normal RXRα function.[1][5][7][9][11][13] Originally discovered at Sanford Burnham Prebys and further developed by Tarrex Biopharma, it represents a first-in-class approach to modulating aberrant RXR signaling in tumors, with early development focused on solid cancers such as colorectal cancer.[3][7][11][13]
