Drug intelligence / Profile preview
TXA11114 is an investigational, small-molecule, indole carboxamide efflux pump inhibitor (EPI) developed by TAXIS Pharmaceuticals. It is designed to combat multidrug-resistant (MDR) *Pseudomonas aeruginosa* infections by inhibiting resistance-nodulation-division (RND) efflux pumps, such as MexAB-OprM, thereby preventing the extrusion of co-administered antibiotics and restoring their intracellular accumulation. In preclinical studies, TXA11114 has been shown to significantly potentiate the antibacterial activity of levofloxacin and other efflux substrates (including tetracyclines, cephalosporins, and chloramphenicol) against MDR clinical isolates. In vivo, the combination of TXA11114 and levofloxacin demonstrated pronounced bacterial clearance in murine thigh and lung infection models, achieving a validated efficacy milestone with a favorable safety and toxicology profile.
