Drug intelligence / Profile preview
TXB4-PD-L1 is a preclinical bispecific fusion protein developed by Ossianix. It consists of the brain-penetrating single-domain shark antibody (VNAR) shuttle TXB4, which targets transferrin receptor 1 (TfR1 / CD71), fused to the anti-PD-L1 monoclonal antibody durvalumab (MEDI4736). By leveraging receptor-mediated transcytosis (or other brain-entry mechanisms) via TfR1, TXB4-PD-L1 is designed to cross the blood-brain barrier (BBB) and deliver the immune checkpoint inhibitor durvalumab into the brain parenchyma. This approach aims to enhance the therapeutic efficacy of PD-L1 blockade in central nervous system (CNS) malignancies, such as primary central nervous system lymphoma (PCNSL) or brain metastases, while minimizing systemic exposure and side effects.
