TXN siRNA is a small interfering RNA (siRNA) designed to silence the expression of the thioredoxin (TXN) gene. Thioredoxin is a critical antioxidant protein that plays a vital role in maintaining cellular redox homeostasis and protecting cells from oxidative stress. In multiple myeloma (MM) research, TXN has been identified as a key inhibitor of ferroptosis, a form of iron-dependent regulated cell death. By utilizing TXN siRNA to knockdown TXN expression, researchers have demonstrated increased lipid peroxidation and inhibited cell proliferation in MM cell lines, such as RPMI8226 and MM.1S. This suggests that the SLC7A11-IRF1-TXN axis serves as a defensive pathway against ferroptosis in cancer cells. Consequently, TXN siRNA serves as both a research tool to elucidate these pathways and a potential precursor to therapeutic strategies aimed at sensitizing resistant multiple myeloma cells to ferroptotic cell death.
