TXN10128 is an orally available small molecule inhibitor of ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1). By inhibiting ENPP1, TXN10128 prevents the degradation of 2'3'-cGAMP in the tumor microenvironment, thereby restoring activation of the STING pathway. This leads to enhanced innate immune activation against tumors and has shown synergistic anti-tumor effects when combined with immune checkpoint inhibitors in preclinical models. The drug demonstrates high selectivity for ENPP1 over related enzymes and favorable pharmacokinetic and toxicological profiles in animal studies. It is being developed primarily for oncology indications such as solid tumors and pancreatic cancer[1][2][4].
