Overview

TY-1054 is an orally bioavailable small-molecule inhibitor of the Transcriptional Enhanced Associate Domain (TEAD) transcription factors, which are key downstream effectors of the Hippo signaling pathway. Developed by TYK Medicines, the compound is designed to disrupt the interaction between TEAD and its co-activators, YAP (Yes-associated protein) and TAZ, thereby suppressing the expression of genes that drive cell proliferation, survival, and epithelial-to-mesenchymal transition. Dysregulation of the Hippo pathway, often resulting from NF2 loss-of-function mutations or YAP/TAZ overexpression, is a significant driver in various malignancies, including mesothelioma and schwannoma. TY-1054 is currently undergoing Phase I/II clinical evaluation for the treatment of advanced solid tumors, including malignant mesothelioma, non-small cell lung cancer, and other Hippo-mutated malignancies.
