Overview

TY-302 is a potent, selective oral small molecule inhibitor of cyclin-dependent kinase 4 and 6 (CDK4/6), developed by TYK Medicines for the treatment of advanced solid tumors, including hormone receptor-positive/human epidermal growth factor receptor 2-negative (HR+/HER2-) advanced breast cancer and metastatic castration-resistant prostate cancer. TY-302 acts by inhibiting CDK4/6, leading to suppression of retinoblastoma protein (Rb) phosphorylation, G1 phase cell cycle arrest, and inhibition of tumor cell proliferation. It is structurally related to palbociclib but has been modified for improved safety. Clinical development includes Phase II trials in breast cancer with plans for registrational studies; it is also being investigated in ovarian cancer (Phase I), acute myeloid leukemia, melanoma, lung cancer, head and neck tumors, and prostate cancer[1][3][4][5][6].
