TY-4028 is an orally bioavailable, small molecule tyrosine kinase inhibitor (TKI) developed by Zhejiang Tongyuan Kang Medicine (also known as TYK Medicines). It is specifically designed to target epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 2 (HER2) harboring exon 20 insertion mutations. These specific genetic alterations are typically resistant to first- and second-generation TKIs due to structural changes in the kinase domain that limit the therapeutic window between mutant and wild-type receptors. TY-4028 aims to selectively inhibit these mutants, thereby blocking downstream signaling pathways such as PI3K/AKT and MAPK/ERK that drive tumor cell proliferation and survival. The compound is currently undergoing Phase 1/2 clinical evaluation to determine its safety, tolerability, and preliminary efficacy in patients with advanced non-small cell lung cancer (NSCLC).
