Overview

**TYK-00540 + enzalutamide** is an investigational combination therapy for advanced/metastatic solid tumors, specifically metastatic castration-resistant prostate cancer (mCRPC). TYK-00540 is a novel, selective small molecule inhibitor targeting cyclin-dependent kinase 2, 4, and 6 (CDK2/4/6), developed by TYK Medicines, intended to inhibit tumor cell growth and overcome resistance to other CDK inhibitors. Enzalutamide (brand name Xtandi) is an approved, second-generation nonsteroidal antiandrogen that acts as an antagonist of the androgen receptor, blocking androgen signaling and inhibiting prostate cancer progression. The combination aims to enhance antitumor efficacy by simultaneously targeting cell cycle progression (via CDK2/4/6 inhibition) and androgen signaling (via androgen receptor inhibition) in patients with mCRPC who have progressed on prior hormonal therapies.
