Tylodipine is a novel, third-generation dihydropyridine calcium channel blocker (CCB) developed by Xuanzhu Biopharmaceutical, a subsidiary of Sihuan Pharmaceutical, for the treatment of essential hypertension. It acts as a potent and selective antagonist of L-type voltage-gated calcium channels (Cav1.2) located in vascular smooth muscle. By inhibiting the influx of calcium ions, tylodipine promotes peripheral vasodilation, thereby reducing systemic vascular resistance and lowering blood pressure. As a third-generation CCB, it is designed to provide a long-lasting antihypertensive effect with a stable pharmacokinetic profile, aiming to minimize common side effects associated with earlier CCBs, such as reflex tachycardia and peripheral edema. The drug is currently in late-stage clinical development in China.
