Overview

Tyloxapol is a nonionic liquid polymer of the alkyl aryl polyether alcohol type used primarily as an inhaled surfactant to aid in the liquefaction and removal of mucopurulent (mucus and pus-containing) bronchopulmonary secretions. It is administered via nebulized solution or oxygen stream for respiratory conditions such as chronic bronchitis and has also been investigated for use in cystic fibrosis and pediatric indications. Tyloxapol acts by reducing surface tension in airway secretions (secretolytic action), dissolving viscous coatings on mucosae to enhance ciliary activity and facilitate expectoration. Additionally, when administered systemically (e.g., intraperitoneally), it inhibits plasma lipoprotein lipase activity—blocking triglyceride breakdown—which has been exploited experimentally to induce hyperlipidemia in animal models. Tyloxapol also exhibits anti-inflammatory effects through inhibition of NF-kappa-B signaling pathways[1][2][3][4].
