Overview

This autologous type-1 polarized dendritic cell (αDC1) vaccine is an experimental personalized immunotherapy developed by Walter J. Storkus at the University of Pittsburgh. The vaccine is produced by harvesting a patient's own dendritic cells and maturing them into a type-1 polarized state, which is optimized for secreting high levels of IL-12p70 to promote potent Th1 and cytotoxic T-lymphocyte (CTL) responses. These cells are loaded with a cocktail of six peptides derived from tumor blood vessel antigens (TBVA) to specifically target the tumor's supporting vasculature in addition to the melanoma cells themselves. It has been primarily investigated in Phase 2 clinical trials for the treatment of metastatic or unresectable melanoma, often in combination with the tyrosine kinase inhibitor dasatinib to overcome immune dysfunction.
