typhoid conjugate vaccine (Zydus Lifesciences Limited)

ZyVac-TCV is a typhoid conjugate vaccine (TCV) developed and manufactured by Zydus Lifesciences (formerly Cadila Healthcare) for the prevention of typhoid fever caused by *Salmonella typhi*. The vaccine is composed of the purified Vi capsular polysaccharide of *S. typhi* covalently conjugated to a tetanus toxoid carrier protein. This conjugation transforms the polysaccharide antigen from a T-cell independent to a T-cell dependent antigen, which significantly enhances immunogenicity, induces immunological memory, and provides longer-lasting protection compared to unconjugated Vi polysaccharide vaccines. It is particularly effective in infants and young children, for whom unconjugated vaccines are typically ineffective. ZyVac-TCV is administered as a single 0.5 mL intramuscular dose and is indicated for individuals aged 6 months to 65 years. The vaccine received World Health Organization (WHO) prequalification in 2024, facilitating its procurement by international agencies for global immunization programs.
