Overview

TYRA-430 is an oral, investigational small molecule inhibitor that selectively targets fibroblast growth factor receptor 4 (FGFR4) and fibroblast growth factor receptor 3 (FGFR3). It is the first reversible FGFR4/3 inhibitor designed to overcome resistance mutations in FGF19-driven cancers, particularly hepatocellular carcinoma (HCC), as well as other advanced solid tumors with activating FGF/FGFR pathway aberrations. Preclinical studies have shown that TYRA-430 exhibits superior potency compared to existing multi-kinase and covalent FGFR4 inhibitors in both *in vitro* and *in vivo* models of HCC and gastric cancer. The drug has demonstrated activity against known FGFR4 resistance mutations. Developed by Tyra Biosciences, it is currently being evaluated in a Phase 1 clinical trial for advanced hepatocellular carcinoma and other solid tumors.
