The Tyrosinase A2 peptide vaccine is an investigational peptide-based cancer immunotherapy developed by Cliniques Universitaires Saint-Luc (Université Catholique de Louvain) in Brussels, Belgium. It is designed as a synthetic peptide vaccine targeting cutaneous melanoma. The active ingredient is the Tyrosinase.A2 peptide (epitope sequence: YMDGTMSQV), which is derived from the melanoma differentiation antigen tyrosinase and presented by HLA class I (HLA-A2) molecules on the surface of cancer cells. The vaccine aims to stimulate an HLA-A2-restricted cytolytic T lymphocyte (CTL) response against tyrosinase-expressing melanoma cells. It was evaluated in a Phase 2 clinical trial (NCT00365937) in disease-free, HLA-A2-positive cutaneous melanoma patients at high risk of relapse, administered either alone or in combination with adjuvants such as Montanide ISA 51 and/or IMP321. The trial was terminated due to regulatory requirements from the peptide supplier, and development of the vaccine has been discontinued.
