Tyrosinase-related protein 1 (TYRP1) is a melanosomal enzyme and member of the tyrosinase family, primarily expressed in melanocytes. It plays a key role in the biosynthesis of melanin, the pigment responsible for skin, hair, and eye color. While its precise catalytic function in human melanocytes is less clear than that of tyrosinase itself, TYRP1 is involved in stabilizing tyrosinase and modulating its activity as well as maintaining melanosome structure. Structurally, it contains two zinc ions at its active site (unlike copper ions found in classic tyrosinases), which explains why it does not exhibit typical redox activity associated with tyrosinases[2][3]. Mutations or alterations in TYRP1 are linked to pigmentation disorders such as oculocutaneous albinism type 3[1]. In oncology research, recombinant forms or peptides derived from TYRP1 have been explored as vaccine antigens or immunotherapy targets for melanoma due to their expression on melanocyte-derived tumors[7][9].
