Overview

Tyrosinase-related protein-1 (TRP-1) peptide vaccine is an investigational cancer immunotherapy developed by the National Cancer Institute (NCI). It is a peptide-based biologic vaccine composed of immunodominant synthetic peptides derived from the TRP-1 (also known as gp75) melanocyte differentiation antigen. TRP-1 is a protein highly expressed in melanocytes and melanoma cells, making it a viable target for therapeutic immunization. The vaccine is designed to stimulate a specific T-cell immune response against melanoma-associated antigens to recognize and destroy tumor cells. It has been evaluated in Phase II clinical trials (notably NCT00019383) for the treatment of recurrent or refractory metastatic melanoma, often administered alongside adjuvants or cytokines such as interleukin-2 (IL-2) or granulocyte-macrophage colony-stimulating factor (GM-CSF) to enhance the anti-tumor immune response.
