This is an experimental therapeutic cancer vaccine developed by Memorial Sloan Kettering Cancer Center (MSKCC) for the treatment of HLA-A2 positive patients with stage III or IV melanoma. The vaccine consists of HLA-A2-restricted peptides derived from the melanoma-associated antigens tyrosinase (YMDGTMSQV) and gp100 (IMDQVPFSV, also known as gp209-2M or IMD-Javelin), which are non-covalently complexed with recombinant 70-kD heat-shock protein (HSP70). HSP70 acts as a molecular chaperone and potent adjuvant, facilitating the uptake of the peptides by antigen-presenting cells (APCs) and enhancing their cross-presentation to CD8+ T cells. The vaccine may also include the OVA BiP peptide as a helper component. The goal of the therapy is to induce a specific and robust cytotoxic T-lymphocyte response against tumor cells expressing these antigens.
