Overview

Tyrosine kinase 2 (TYK2) short hairpin RNA (shRNA) is an experimental RNA interference (RNAi) agent designed to silence the expression of the TYK2 gene. In the context of oncology research, specifically breast cancer, TYK2 has been identified as a mechanosensitive metastasis suppressor that preserves epithelial integrity under low extracellular matrix (ECM) stiffness. Depletion of TYK2 via shRNA leads to the nuclear translocation of the transcription factor TWIST1, triggering epithelial-mesenchymal transition (EMT) and multicellular invasion. While TYK2 inhibitors like deucravacitinib are used clinically for autoimmune diseases, experimental use of TYK2 shRNA in triple-negative breast cancer (TNBC) models has demonstrated that loss of TYK2 promotes local invasion and lung metastasis without affecting primary tumor growth. This suggests that TYK2 serves as a fundamental stiffness-governed epithelial defense mechanism.
