Tyrosinyl-amantadine (Tyr-Am) is a synthetic derivative of amantadine designed for the treatment of Parkinson's disease. It is a conjugate of the amino acid L-tyrosine and amantadine, intended to leverage the neuroprotective properties of both components or improve the delivery and potency of amantadine. Preclinical studies in 6-hydroxydopamine (6-OHDA) rat models indicate that Tyr-Am possesses significant antioxidant properties, specifically reducing lipid peroxidation (LPO) and maintaining superoxide dismutase (SOD) activity. It has been shown to improve neuromuscular coordination and cognitive performance (learning and memory) at doses approximately 2.5 times lower than those required for amantadine, while having no significant effect on acetylcholinesterase activity.
