TZ-POR 7e is a tetrazine-functionalized porphyrin derivative designed for use in pretargeted photodynamic therapy (PT-PDT). It serves as a bioorthogonal photosensitizer that utilizes the inverse electron-demand Diels-Alder (IEDDA) reaction to covalently 'click' onto trans-cyclooctene (TCO)-modified targeting vectors, such as monoclonal antibodies, that have been previously localized at a tumor site. This pretargeting strategy is intended to enhance the therapeutic window of photodynamic therapy by allowing the photosensitizer to accumulate specifically at the target tissue while clearing from systemic circulation, thereby reducing off-target phototoxicity. Once localized and activated by light of a specific wavelength (typically in the red or near-infrared spectrum), the porphyrin core of TZ-POR 7e acts as a catalyst to transfer energy to molecular oxygen, generating highly cytotoxic singlet oxygen and other reactive oxygen species (ROS). These species induce localized oxidative damage, leading to tumor cell apoptosis, necrosis, and vascular shutdown.
