TZP-102

TZP-102 is a novel, orally administered, macrocyclic small molecule that acts as a selective agonist of the ghrelin (growth hormone secretagogue) receptor. It was developed as a prokinetic agent for the treatment of gastroparesis, particularly in patients with diabetes mellitus. The drug works by stimulating the ghrelin receptor, which plays a direct role in promoting gastrointestinal motility and accelerating gastric emptying. TZP-102 was evaluated in multiple clinical trials for diabetic gastroparesis and received FDA Fast Track designation due to the significant unmet need in this patient population. Despite initial promise and evidence of symptom improvement, development was ultimately discontinued after mid-stage studies failed to meet primary efficacy endpoints.
