U-359 is a novel synthetic uracil analog that exhibits cytotoxic effects against breast cancer cells and demonstrates potential for overcoming chemotherapy resistance. Its mechanism of action involves inducing apoptosis, characterized by the activation of caspases 8 and 9, and increased levels of cleaved PARP1. Furthermore, U-359 down-regulates the expression of key ATP-binding cassette (ABC) transporters, including ABCB1, ABCG2, and ABCC1, and modulates NF-κB protein levels, which are implicated in multidrug resistance. It also contributes to reversing Taxol resistance by reducing the overexpression of tubulin III and Nlp proteins.
