U-74389G liposomes are a PEGylated liposomal formulation of the 21-aminosteroid U-74389G, a member of the lazaroid family. This formulation is designed to enhance the delivery of U-74389G, a potent inhibitor of lipid peroxidation, to the brain. In preclinical glioblastoma models, it demonstrates dual efficacy by protecting healthy brain tissue from radiation-induced oxidative stress and exerting direct antiproliferative effects on tumor cells. The formulation, characterized by a size of 80-90 nm, was developed to overcome the pharmacokinetic limitations of the parent compound and improve its therapeutic index in treating central nervous system malignancies.
