U1-59

U1-59 is a fully human monoclonal antibody directed against human epidermal growth factor receptor 3 (HER3, also known as ErbB3). Originally co-developed by U3 Pharma and Amgen, and later owned by Daiichi Sankyo following its acquisition of U3 Pharma, U1-59 binds to the extracellular domain of HER3. This binding blocks the interaction of HER3 with its ligands, such as neuregulin-1 (NRG1) or heregulin, thereby preventing receptor heterodimerization with other HER family members (such as HER2 or EGFR) and inhibiting downstream PI3K/Akt signaling pathways. U1-59 has been evaluated in preclinical models for various solid tumors, including breast, lung, pancreatic, and prostate cancers, and has also served as the targeting antibody component in experimental anti-HER3 antibody-drug conjugates (ADCs).
