Overview

U3-1287 + trastuzumab + paclitaxel is an experimental combination therapy for advanced solid tumors and HER2-positive breast cancer. U3-1287 (patritumab, also known as AMG 888) is a fully human monoclonal antibody targeting HER3, aiming to inhibit oncogenic signaling mediated by the HER3 receptor, which plays a critical role in resistance to HER family inhibitors. Trastuzumab is a monoclonal antibody against HER2, used for HER2-overexpressing breast cancer, functioning by blocking HER2-mediated cell growth. Paclitaxel is a microtubule-stabilizing small molecule chemotherapeutic that disrupts mitosis. Together, this combination targets HER2 and HER3 receptors, aiming for synergistic anti-tumor activity by dual inhibition of oncogenic pathways and cytotoxicity. U3-1287 is primarily developed by Daiichi Sankyo/U3 Pharma, trastuzumab by Genentech (Roche), and paclitaxel is produced by various companies.
