U99194 (often supplied as the maleate salt) is a **potent and moderately selective antagonist of the dopamine D3 receptor**, exhibiting approximately 15-30-fold preference for D3 over the D2 receptor, and more than 10,000-fold selectivity over the D4 receptor. It has been widely used in pharmacological research to probe the role of the D3 receptor in neuropsychiatric conditions and drug addiction. In rodents, U99194 shows **biphasic effects on reward and locomotor activity**, likely due to opposing effects at autoreceptors and postsynaptic receptors. It also affects nociception, thermoregulation, anxiety, learning and memory, and modulates behavioral responses to psychostimulants and opioids. U99194 is not approved for clinical use and is mainly used as a research tool[1][3][5][8].
