UA021

UA021 is an oral, first-in-class small-molecule inhibitor of tyrosine-protein kinase 2 (TYK2) being developed by Usynova Pharmaceuticals (also referenced as Yousen Jianheng Biomedical) for the treatment of autoimmune and inflammatory diseases including psoriasis, inflammatory bowel disease, and systemic lupus erythematosus.[1][3][13] As a highly potent and selective inhibitor of allosteric TYK2 variants with good oral bioavailability, UA021 is designed to block TYK2-mediated cytokine signaling pathways (such as IL-12, IL-23, and type I interferons) that drive pathogenic immune activation while potentially minimizing safety liabilities associated with broader JAK inhibition.[1][3] The drug is currently in early clinical development, with Phase 1 single- and multiple-ascending dose studies in healthy volunteers to characterize safety, tolerability, pharmacokinetics, and food effects, and its global highest development phase is reported as Phase 1 for psoriasis.[1][3][6][11][13]
