Overview

UB-221 is a third-generation, humanized IgG1 monoclonal antibody that targets immunoglobulin E (IgE). It is designed to neutralize free IgE with high affinity and inhibit the synthesis of new IgE, mechanisms not fully achieved by earlier anti-IgE therapies such as omalizumab. UB-221 binds both high-affinity (FcεRI) and low-affinity (CD23) IgE receptors, effectively preventing basophil activation and degranulation. Preclinical studies demonstrated rapid reduction in serum IgE levels, while clinical trials in patients with chronic spontaneous urticaria showed durable symptom relief associated with decreased free serum IgE. The drug is being developed primarily for allergic diseases including chronic urticaria, atopic dermatitis, allergic asthma, food hypersensitivity, and allergic rhinitis[1][3][5][7].
