UB-312 is a synthetic peptide-based vaccine designed as an active immunotherapy to stimulate an immune response against pathological forms of alpha-synuclein (α-synuclein). It incorporates a 10-amino-acid fragment from the C-terminal domain of α-synuclein, conjugated to a T-helper peptide using UBITh technology, and is administered intramuscularly. The vaccine induces antibodies that preferentially bind to aggregated and toxic forms of α-synuclein (oligomers and fibrils), which are implicated in the pathogenesis of synucleinopathies such as Parkinson's disease, multiple system atrophy (MSA), and dementia with Lewy bodies. Preclinical studies demonstrated that UB-312 immunization reduces α-synuclein pathology and improves motor function in animal models. In clinical trials, UB-312 has shown safety, high immunogenicity, target engagement in cerebrospinal fluid, reduction of aggregated α-synuclein levels, and improvement in motor experiences for patients with Parkinson’s disease. The drug is being developed by Vaxxinity for disease modification or prevention in synucleinopathies[1][2][3][4][5][6][7][8].
