UB-612 is a multitope protein/peptide subunit vaccine developed for the prevention of COVID-19. It is designed to activate both B-cell and T-cell arms of the immune system by targeting multiple structural antigens of SARS-CoV-2, including the spike (S1-RBD and S2), nucleocapsid (N), and membrane (M) proteins. The vaccine contains a CHO cell–produced spike protein receptor-binding domain fused with a single-chain Fc region, five synthetic peptides representing conserved T helper and cytotoxic T lymphocyte epitopes from N, M, and S2 proteins, as well as an extrinsic HLA class II epitope derived from measles virus fusion protein to enhance immunogenicity. Adjuvants such as CpG oligonucleotides and aluminum phosphate are included to further boost immune response. UB-612 has demonstrated robust induction of neutralizing antibodies against multiple SARS-CoV-2 variants—including Alpha, Beta, Gamma, Delta, Omicron—and long-lasting B-cell and broad T-cell immunity in preclinical models and clinical trials. It is being developed for both primary immunization and heterologous booster use[1][2][3][4][5][6].
