Overview

UB-VV400 + rapamycin is an investigational **combination therapy** comprising UB-VV400, an in vivo CD22-directed CAR-T cell therapy, and rapamycin, an oral mTOR inhibitor. UB-VV400 is engineered to target CD22, a clinically validated B-cell antigen expressed in most B-cell malignancies, and incorporates a synthetic cytokine system (RACR2) to enhance CAR-T cell survival and potency. Rapamycin (also known as sirolimus) is used for its antiproliferative and immunomodulatory properties; it inhibits the mammalian target of rapamycin (mTOR) pathway, modulating cellular growth, proliferation, and survival. The combination is intended to address relapsed or refractory large B-cell lymphomas (LBCL), including DLBCL, high-grade lymphoma, follicular lymphoma Grade 3B, and primary mediastinal B-cell lymphoma, particularly in populations exposed to prior CD19-directed CAR-T therapy or those who are CAR-T naive. It is in early-stage clinical development focusing on safety, pharmacokinetics, and efficacy optimization[1][3][4][5].
