UBCS060 is a synthetic small molecule pyrrolo[1,2-a]quinoxaline derivative that serves as a regioisomer negative control for the Sirtuin 6 (SIRT6) activator UBCS039. It exhibits very low affinity for the SIRT6 enzyme and lacks the ability to stimulate SIRT6-dependent deacetylation of histone H3 at the K9 and K56 positions. In pharmacological research, UBCS060 is utilized to demonstrate that the biological effects observed with SIRT6 activators—such as the induction of autophagic flux, reactive oxygen species (ROS) accumulation, and autophagy-related cell death in various human cancer cell lines—are specific to the activation of the SIRT6 enzyme rather than non-specific effects of the chemical scaffold. Consequently, UBCS060 does not affect cancer cell proliferation, cell cycle progression, or apoptosis.
