DN-UBE2C is a dominant negative mutant form of the ubiquitin-conjugating enzyme E2 C (UBE2C). This mutant is used in experimental research to functionally inhibit the wild-type UBE2C protein, which plays a key role in ubiquitin-mediated protein degradation, cell cycle progression, and cancer cell survival. In studies, DN-UBE2C has been shown to reduce the growth rate and anchorage-independent growth of cancer cells, and sensitize breast cancer cells to radiation, chemotherapy (e.g., doxorubicin), and hormone-blocking agents (e.g., tamoxifen, letrozole). Mechanistically, inhibiting UBE2C affects cell cycle progression, induces cell cycle arrest (particularly at the G2/M phase), and promotes apoptosis, making it a potential candidate for cancer therapy, primarily as a preclinical research tool rather than a clinical therapeutic[2].
