Ublituximab + umbralisib (TGR-1202) + bendamustine is a combination therapy under investigation for the treatment of relapsed or refractory B-cell non-Hodgkin lymphoma (NHL), including diffuse large B-cell lymphoma (DLBCL), follicular lymphoma (FL), small lymphocytic lymphoma (SLL), and marginal zone lymphoma (MZL)[1][4][5]. Ublituximab is a glycoengineered monoclonal antibody targeting CD20 on B cells to induce immune-mediated lysis. Umbralisib (TGR-1202) is an orally available phosphoinositide 3-kinase delta inhibitor that blocks PI3Kδ signaling in malignant B cells. Bendamustine is an alkylating agent that crosslinks DNA and induces cell death[4][6]. The combination leverages non-overlapping mechanisms to enhance anti-tumor activity in patients with advanced or refractory NHL who may not be eligible for high-dose chemotherapy or stem cell transplant[1][4].
