U2-CHOP is an investigational chemoimmunotherapy regimen consisting of ublituximab, umbralisib, and CHOP chemotherapy (cyclophosphamide, doxorubicin, vincristine, and prednisone). Ublituximab is a glycoengineered anti-CD20 monoclonal antibody designed with low fucose content to enhance antibody-dependent cellular cytotoxicity (ADCC). Umbralisib is a dual inhibitor of PI3K-delta and casein kinase 1 (CK1) epsilon. This combination is being evaluated in a Phase Ib/II clinical trial led by the University of Alabama at Birmingham for the treatment of previously untreated mantle cell lymphoma (MCL). The regimen typically involves an induction phase with U2-CHOP followed by a maintenance phase with the U2 (ublituximab and umbralisib) combination. The multi-targeted approach aims to deplete B-cells, inhibit B-cell receptor signaling, and induce cytotoxic DNA damage.
