Overview

Ubv-A is an engineered ubiquitin variant developed by Integrated DNA Technologies (IDT) to enhance the efficiency of homology-directed repair (HDR) during genome editing. It functions as an inhibitor of p53-binding protein 1 (53BP1), a protein that normally promotes non-homologous end joining (NHEJ) and suppresses HDR at DNA double-strand breaks. By blocking 53BP1 recruitment, Ubv-A shifts the cellular repair machinery toward the HDR pathway, allowing for more precise genetic modifications. Ubv-A was identified through a two-hybrid screen for variants with improved binding affinity for 53BP1 compared to earlier variants like i53, and it is optimized for delivery as a purified protein during ex vivo nucleofection.
