UBX0101 is a small molecule senolytic drug developed for the treatment of musculoskeletal diseases, with an initial focus on osteoarthritis (OA) of the knee. It acts as an inhibitor of the p53/MDM2 protein-protein interaction by binding to Mouse double minute 2 homolog (MDM2), which raises Tumor protein p53 (p53) levels and induces apoptosis in senescent cells. The elimination of these cells is believed to create a pro-regenerative environment in joint tissues and potentially reduce pain and improve function. Preclinical studies demonstrated that UBX0101 could selectively clear senescent cells from cartilage and synovium, leading to attenuation of OA progression in animal models. Clinical trials showed that intra-articular administration was well-tolerated but did not meet primary efficacy endpoints in Phase II studies for knee OA[1][2][4][5][8].
