Overview

UC-781 is a thiocarboxanilide non-nucleoside reverse transcriptase inhibitor (NNRTI) developed as a topical microbicide for the prevention of HIV transmission. It acts by binding with high affinity to HIV-1 reverse transcriptase, inhibiting both DNA synthesis and pyrophosphorolysis catalyzed by wild-type and certain mutant forms of the enzyme. This inhibition can restore the chain-terminating activity of zidovudine (AZT) against AZT-resistant virus. UC-781 demonstrates potent antiviral and virucidal activity in vitro, is stable under low pH and various temperature conditions, and has been formulated into gels for vaginal or rectal application. Clinical trials have shown that topical administration is safe with no significant adverse events or systemic absorption detected[1][2][3][4][5][6].
